The defense and the prosecution have rested and the jury is filing into the jury room to decide if a young Spanish-American is guilty or innocent of murdering his father. What begins as an open and shut case of murder soon becomes a mini-drama of each of the jurors' prejudices and preconceptions about the trial, the accused, and each other. Based on the play, all of the action takes place on the stage of the jury room.

Al termine dell'ultima udienza di un processo per omicidio, 12 giurati si chiudono in camera di consiglio. L'intera pellicola si sviluppa dentro questo ambiente claustrofobico, mettendo a confronto le personalità, gli stati d'animo, i conflitti culturali ed etnici dei 12 protagonisti chiamati a giudicare della sorte di un uomo. Inizialmente, tutti i giurati, tranne uno sono convinti della colpevolezza dell'imputato: lentamente, dal continuo confronto tra le opinioni dei giurati (un confronto che spesso trascende nel contrasto verbale e fisico) soprattutto tra Fonda e l'altrettanto grande Balsam che interpreta il presidente della giuria, emergono tutte le contraddizioni dei testimoni, non adeguatamente valutate dall'avvocato difensore d'ufficio...
